As an early career investigator, I hold a K08 from the NINDS entitled Investigation of Histone H3 Post-Translational Modifications in Pediatric Brainstem Glioma. Through this project I investigate the epigenetic landscape of pediatric high grade and brainstem glioma, as well as molecular mechanisms of pediatric gliomagenesis using patient derived tissue, primary cell lines, and a mouse xenograft model of disease. Specifically, my group investigates the effects of the Histone H3 mutations (H3K27M and H3G34V) on gene transcription and protein post-translational modifications to promote oncogenesis. We use these data to then identify novel, rational therapeutic vulnerabilities and explore the use of epigenetic targeted drugs, including EZH2 and BRD inhibition. My group also works on identifying clinically detectable biomarkers of pediatric brain tumors, and was the first to report detection of both tumor related proteins, and, later, tumor derived mutant DNA (H3K27M and H3G34V mutations). We are currently establishing a study to longitudinally collect CSF, blood and urine from children with brain tumors in order to determine utility of a liquid biopsy approach for stratification to targeted therapies and monitoring of disease. Lastly, my group is interested in novel methods of drug delivery across the blood brain barrier to improve penetrance and delivery to tumor cells. To this end, we are working to establish a study using low intensity pulsed ultrasound to disrupt the blood brain barrier in our pediatric glioma mouse model, with the goal to establish the requisite pre-clinical data necessary to translate this approach into a clinical trial in children.
