Researchers in this study wanted to understand whether genetic ancestry influences normal breast tissue structure, and how these differences might be connected to breast cancer risk. One important feature they studied is terminal duct lobular unit (TDLU) involution, which is the natural shrinking of breast tissue structures as women age. Less involution—meaning more remaining glandular structures—has been linked to a higher risk of breast cancer.
To investigate this, researchers analyzed normal breast tissue samples from a large group of women and used genetic data to estimate each participant’s ancestry. They then measured features of breast tissue, including the number and size of TDLUs, and compared these characteristics across individuals.
They found that women with a higher proportion of African genetic ancestry tended to have less TDLU involution, meaning more glandular structures remained in the breast. These differences were still present even after accounting for other factors such as age, body mass index, and reproductive history.
Overall, the study suggests that normal differences in breast tissue structure related to genetic ancestry may help explain variations in breast cancer risk across populations.
